Arp2 Antibody (E-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects Arp2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Arp2 (E-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Arp2 protein plays a crucial role in actin polymerization, which is essential for numerous cellular functions such as chemotaxis, cell migration, cell adhesion, and platelet activation. Arp2 is a key component of the Arp2/3 complex, which facilitates the nucleation of new actin filaments and promotes the formation of a branched actin network. This process is vital for maintaining cellular structure and enabling dynamic changes in cell shape and movement. The Arp2/3 complex operates by binding to the sides of existing actin filaments or the pointed ends, thereby catalyzing the growth of new filaments in the barbed direction. The activity of the Arp2/3 complex is regulated by small GTPases from the Rho family, such as Cdc42, which, upon activation, recruits WASP proteins that further stimulate the complex to initiate the formation of lamellipodia and filopodia. This intricate regulation underscores Arp2′s importance in orchestrating the cytoskeletal dynamics necessary for various cellular processes.

I am using Arp2 (E-12): sc-166103 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Rated 4 out of 5 by from Works well on westerns, although not for IF or IPThe Arp2 antibody (E-12) detected the expected ~45Kda band and a much weaker ~70kDa band on westerns with total lysates of human tissue culture cells. There was no specific signal in the IF with mouse or human cells and the antibody also did not work for IP from cytoplasmic lysates of human tissue culture cells

Rated 4 out of 5 by from No cross species reactivity against S. cerevisiaeWe tested Antibody Arp2 (E-12) against S. cerevisiae Arp2 subunit as shown in the picture. Bovine Arp2/3 complex was loaded as a positive control and Arp2 was very well blotted. Same amount of budding yeast (S. cerevisiae) Arp2/3 complex was loaded, but Arp2 was not detected. Alternatively, Arp3 may be weakly detected (1:500 dilution, when used for the 2nd time, the signal was too low to be seen). Cell lysate of budding yeast was tested as well and nothing was detected. (sc: budding yeast Arp2/3 complex; cl: cell lysate of budding yeast; bt: bovine Arp2/3 complex).

Rated 3 out of 5 by from OK to use if Arp2 is abundantly expressedI received a free sample about two months ago as a replacement of a rabbit polyclonal that was discontinued. Among the many anti-Arp2 antibodies I have checked E-12 was the only one yielding a single band at the appropriate size in western blotting of both mouse and human endothelial cell whole cell lysates (albeit the signal was very low in the mouse sample). However, I bought the same antibody in the largest size and this time I did not get any signal on the same amount of material transferred to PVDF-FL in the same conditions. The detection system was NIR fluorescence. I have not checked this antibody with chemiluminescence.
