EWS Antibody (C-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects EWS in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). EWS Antibody (C-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. EWS is a crucial nuclear RNA-binding protein that plays a significant role in gene regulation and cellular processes. Notably, EWS gene is often involved in chromosomal translocations that lead to the formation of fusion proteins with transcription factors, such as ATF-1, which are implicated in human neoplasias. In Ewing sarcoma, EWS protein is fused to the DNA-binding domain of ETS transcription factors, including Fli-1, Erg, ETV1, E1AF, and FEV, resulting in the EWS/Fli-1 chimeric protein. This fusion protein acts as a potent transcriptional activator, enhancing the expression of target genes and promoting cell transformation, which is critical for tumorigenesis. EWS protein contains two functional regions: an amino-terminal region (domain A) that has limited transactivation activity but can efficiently induce transformation when fused to Fli-1, and a distal region (domain B) that exhibits transactivation activity but is less effective in promoting transformation when associated with Fli-1. Understanding the functional dynamics of EWS and its interactions is essential for elucidating its role in cancer biology and developing targeted therapeutic strategies.

I am looking at C9 antibody for EWSR1 and also for EWS-FLI1 fusion protein detection in Ewing Sarcoma. it appears this Ab detects AA 2-19 but the first citation states it is detecting an epitope AA 431 to 490. Conspicuously, ab G5 is for AA 2-19.

Can the peptide sc-48404 P be used as a positive control in Western Blot?

Thank you for your question. The blocking peptide sc-48404 P is a short synthetic peptide intended for use as a negative control. It is not appropriate for use as a positive control in Western Blot.
