Heme Oxygenase 1 Antibody (A-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Heme Oxygenase 1 (HO-1) of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-HO-1 antibody (A-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HO-1, also known as HSP 32, plays a crucial role in cellular defense mechanisms by catalyzing the degradation of heme into biliverdin, carbon monoxide, and free iron, thereby providing antioxidant protection and regulating inflammation. The production of carbon monoxide by HO-1 is particularly significant as a signaling molecule that can modulate various physiological processes, including vasodilation and neurotransmission. HO-1 is highly inducible in response to stressors such as heavy metals, hormones, and oxidative stress, making HO-1 a key player in the cellular response to injury and inflammation. In contrast, Heme Oxygenase 2 is constitutively expressed in mammalian tissues and is primarily involved in carbon monoxide production in the brain, where carbon monoxide is thought to function as a neurotransmitter. Understanding HO-1 function is essential for exploring potential therapeutic applications in conditions characterized by oxidative stress and inflammation.

Thank you for your question. Heme Oxygenase 1 Antibody (A-3) is only recommended for detection of Heme Oxygenase 1 of human origin by WB, IP, IF, IHC(P) and ELISA. For use with human samples you may want to opt for Anti-Heme Oxygenase 1 Antibody (A-3): sc-136960 or Anti-Heme Oxygenase 1 Antibody (F-4): sc-390991.

Will this HO-1 antibody react with rat? I am trying to perform IHC (F) on rat brain sections.

Thank you for your question. Heme Oxygenase 1 Antibody (A-3) is only recommended for detection of Heme Oxygenase 1 of human origin by WB, IP, IF, IHC(P) and ELISA. Based on our lab's tests we do not recommend it for rat cells or tissues. Instead you could opt for Heme Oxygenase 1 Antibody (F-4): sc-390991, which is recommended for detection of Heme Oxygenase 1 of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA.

How can Heme Oxygenase 1 (A-3): sc-136960 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. Heme Oxygenase 1 (A-3): sc-136960 is currently available conjugated to phycoerythrin (sc-136960 PE), fluorescein (sc-136960 FITC), Alexa Fluor® 488 (sc-136960 AF488) or Alexa Fluor® 647 (sc-136960 AF647)
