MBP Antibody (F-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects MBP protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). MBP Antibody (F-6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Myelin basic protein (MBP) plays a crucial role as the primary extrinsic membrane protein of central nervous system myelin, essential for the formation and maintenance of the myelin sheath that insulates nerve fibers, thereby facilitating efficient signal transmission. MBP (F-6) antibody enables researchers to study the phosphorylation of MBP at Threonine 125, a complex regulatory mechanism that significantly influences myelin sheath stability. This phosphorylation is modulated by mitogen-activated protein kinases during myelinogenesis and in demyelinating diseases such as multiple sclerosis. Notably, MBP phosphorylation responds to high-frequency stimulation of the alveus, the myelinated output fibers of the hippocampus, but not to low-frequency stimulation. During heightened neuronal activity, calcium influx activates axonal nitric oxide synthase, leading to the production of intercellular messengers like nitric oxide and superoxide, which regulate MBP phosphorylation through MAPK pathways. This intricate regulation underscores MBP′s importance in both normal and pathological states of the nervous system, making anti-MBP antibody (F-6) a valuable tool for research in neurobiology and related fields.

Can this antibody be used for FACS of rat oligodendrocytes?

is this antibody suitable for IF staining of human oligodendrocytes?

Can this antibody be used successfully to stain mouse tissue in IHC(P)?

Thank you for your question. Yes, this antibody is appropriate for IHC of paraffin-embedded mouse tissue. In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is currently available conjugated to HRP, PE, FITC, AF488 and AF647.

Cryostat-cut brain slices were incubated free-floating overnight with this antibody (1:200 in PB + 0.3% Triton X-100), followed by 2-h incubation with a fluorescently-labelled secondary antibody (red signal in figure), while nuclei were counterstained with DAPI (blue signal in figure).
