NQO1 Antibody (A180) is a mouse monoclonal IgG1 kappa light chain antibody that detects NQO1 protein of human, mouse, and, to a lesser extent, rat origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. NQO1 (A180) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NQO1, also known as NAD(P)H:quinone oxidoreductase 1, plays a crucial role in cellular defense by catalyzing the reduction of quinones to hydroquinones, thereby detoxifying harmful compounds and protecting cells from oxidative stress, cytotoxicity, and mutagenicity. This enzymatic activity is particularly important in cancer, as many tumors overexpress NQO1, which serves as an obligate two-electron reductase that not only deactivates toxins but also activates bioreductive anticancer drugs. NQO1 is a 274 amino acid protein ubiquitously expressed across various tissues, although expression levels can vary significantly. NQO1 gene expression is tightly controlled and can be induced by factors such as xenobiotics, antioxidants, heavy metals, and radiation, with the antioxidant response element (ARE) in the NQO1 promoter playing a pivotal role in this process. ARE activation by compounds like tert-butylhydroquinone depends on the PI3-kinase pathway, which is upstream of the transcription factor Nrf2. Various transcription factors, including Nrf2, c-Jun, Nrf1, Jun-B, and Jun-D, bind to the ARE to regulate NQO1 expression, while Maf-Maf homodimers and Maf-Nrf2 heterodimers may negatively regulate this transcriptional activation. This intricate regulation of NQO1 is essential for maintaining cellular homeostasis and responding to oxidative stress, making anti-NQO1 antibody (A180) a valuable tool for researchers studying these critical biological processes.
