NQO1 Antibody (A180) is a mouse monoclonal IgG1 kappa light chain antibody that detects NQO1 protein of human, mouse, and, to a lesser extent, rat origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. NQO1 (A180) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NQO1, also known as NAD(P)H:quinone oxidoreductase 1, plays a crucial role in cellular defense by catalyzing the reduction of quinones to hydroquinones, thereby detoxifying harmful compounds and protecting cells from oxidative stress, cytotoxicity, and mutagenicity. This enzymatic activity is particularly important in cancer, as many tumors overexpress NQO1, which serves as an obligate two-electron reductase that not only deactivates toxins but also activates bioreductive anticancer drugs. NQO1 is a 274 amino acid protein ubiquitously expressed across various tissues, although expression levels can vary significantly. NQO1 gene expression is tightly controlled and can be induced by factors such as xenobiotics, antioxidants, heavy metals, and radiation, with the antioxidant response element (ARE) in the NQO1 promoter playing a pivotal role in this process. ARE activation by compounds like tert-butylhydroquinone depends on the PI3-kinase pathway, which is upstream of the transcription factor Nrf2. Various transcription factors, including Nrf2, c-Jun, Nrf1, Jun-B, and Jun-D, bind to the ARE to regulate NQO1 expression, while Maf-Maf homodimers and Maf-Nrf2 heterodimers may negatively regulate this transcriptional activation. This intricate regulation of NQO1 is essential for maintaining cellular homeostasis and responding to oxidative stress, making anti-NQO1 antibody (A180) a valuable tool for researchers studying these critical biological processes.

Dear team, My advisor buy this code NQO1 (A180): sc-32793 in IHC-P application . I would like to ask in Positive control for tonsil Where NQO1 will it be attach at squamous epithelium or vessel ?

Asked by: Yanisorn
Thank you for your question. NQO1 is expressed in the cytoplasm, so cytoplasmic staining is expected. I do not have any information regarding specific attachment in tonsil, so I advice you to look in the primary literature for further information. https://www.uniprot.org/uniprot/P15559

We tested the NQO1 A180 antibody on rat spinal cord protein extract and see no band at 45 ug protein. The same sample showed strong band of HO-1 as well as Iba-1. Can you help us to resolve our problem? What kind of secondary antibody should I use?

Asked by: Sungreace
Thank you for your question. Please contact our Technical Service Department or your local distributor for assistance in troubleshooting this antibody. We are happy to help!

I want to perform a double stain in my samples, how can I do it with only mouse monoclonal antibodies?

Asked by: Cweed
We recommend using conjugated primary antibodies. This product, sc-32793, is available directly conjugated to FITC, AF488, and AF 647.

Rated 2 out of 5 by from NQO1 Slight signal for WBIt showed a slight signal at the expected molecular weight. It shows a more evident nonspecific signal at 55 kDa. I use it at a dilution of 1:1000 on lisates from murine hepatocytes. It seems not so well for my WB.
