Sp1 Antibody (1C6) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 609-627 mapping to an internal domain of human-origin Specificity Protein 1 (Sp1), a transcription factor involved in the regulation of numerous genes. Sp1 monoclonal antibody (1C6) reacts with mouse, rat, and human species and is suitable for applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry on paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Sp1, also known as Transcription Factor Sp1, is a zinc finger protein that binds to GC-rich motifs in DNA, commonly referred to as GC boxes, playing a crucial role in gene expression, cell growth, differentiation, and apoptosis. Initially identified as a factor that activates transcription from the SV40 promoter by binding to multiple GC boxes in the 21-base pair repeats, Sp1′s DNA-binding specificity is conferred by three Cys2His2-type zinc finger motifs. Phosphorylation of Sp1 can modulate activity and DNA-binding affinity, affecting the transcription of target genes. Located predominantly in the nucleus, Sp1′s localization is essential for function as a transcriptional regulator. Dysregulation of Sp1 has been implicated in various diseases, including cancer, highlighting importance in maintaining normal cellular functions. Sp1 monoclonal antibody (1C6) enables researchers to study the expression and function of Sp1 in different biological contexts, aiding in the understanding of its role in health and disease.

Thank you for your question. We recommend using our mouse monoclonal, Sp1 Antibody (1C6): sc-420 for ChIP. We recommend purchasing the more concentrated TransCruz reagent for ChIP applications (sc-420 X, 200 µg/0.1 ml). This antibody has been published for use in ChIP applications (e.g., PMID's: 16901903, 17059557 and 20351094).

Can this mouse monoclonal antibody be used for IHC staining of mouse tissue?

Yes. We recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody for Sp1 is available conjugated to agarose (sc-420 AC), 500 µg/0.25 ml agarose in 1 ml, for IP; HRP (sc-420 HRP), 200 µg/ml, for WB, IHC(P) and ELISA; phycoerythrin (sc-420 PE), fluorescein (sc-420 FITC), Alexa Fluor® 488 (sc-420 AF488) or Alexa Fluor® 647 (sc-420 AF647), 200 µg/ml, for IF, IHC(P) and FCM

What is the difference between sc-420 and sc-420 P?

Thank you for your inquiry. sc-420 is our standard unconjugated Sp1 (1C6) monoclonal antibody. sc-420 P is the blocking peptide matching this antibody that can be used as a negative control in combination with the antibody. Rated 1 out of 5 by from Did not work for meThis did not work for me on Western blot with 50 ug of HeLa lysate at 1:250 dilution. I saw some weak non-specific bands and nothing on the size range of Sp1. It might work for other applications or with other tissues/cells. This is all I tested it with.
