ZW10 Activators

ZW10, also known as Zeste White 10 or mitotic checkpoint protein ZW10, is a critical component of the spindle assembly checkpoint (SAC), a safety mechanism ensuring correct chromosome segregation during mitosis. This protein functions as a signal transducer for the SAC by localizing to kinetochores-structures on chromosomes where spindle fibers attach-during mitosis. ZW10 is involved in delaying the onset of anaphase until all chromosomes are properly aligned and attached to the spindle, thus blocking aneuploidy, which can lead to cancer if left unchecked.

The protein interacts with other checkpoint proteins, such as Rod and Zwilch, forming the RZZ complex, which is essential for the recruitment of dynein-dynactin to kinetochores and the generation of the checkpoint signal. When chromosomes are improperly attached, ZW10 helps inhibit the activity of the anaphase-promoting complex/cyclosome (APC/C), a ubiquitin ligase that triggers the progression into anaphase by targeting specific proteins for degradation.Besides its well-established role in mitosis, ZW10 has been implicated in other cellular processes, such as membrane trafficking and the regulation of centrosome functions. Mutations or dysregulation of ZW10 have been associated with various types of cancer due to its role in maintaining genomic stability.
